A carbon 60 fullerene is a polyhedral carbon structure composed of around 60 carbon atoms in pentagon and hexagon configuration. They are named after Buckminster Fuller because of structural resemblance to geodesic domes (Chemid, 2015). The carbon 60 fullerene has a log Kow = 6.67 which means that is very lipophilic and is not very soluble in aqueous environments (HSDB, 2013). Literature suggesting there is toxicity from carbon 60 exposure is nearly exclusively focused on water based formulations of carbon 60 (HSDB, 2013). Carbon 60 does not perpetuate Fenton reactions which would produce oxygen radicals in the presence of metal ions (HSDB, 2013). Fullerenes are powerful antioxidants, reacting readily and at a high rate with free radicals, which are often the cause of cell damage or death. Fullerenes hold great promise in health and personal care applications where prevention of oxidative cell damage or death is desirable, as well as in non-physiological applications where oxidation and oxygen free-radical processes are destructive including but not limited to, food spoilage, plastics deterioration, and metal corrosion (HSDB, 2013).

Carbon 60 in a fullerene is 172 times better at scavenging oxygen radicals than Vitamin C. The antioxidant property is based on the fact that fullerenes possess large amount of conjugated double bonds and low lying lowest unoccupied molecular orbital which can easily take up an electron, making an attack of radical species highly possible. It has been reported that up to 34 radicals have been added onto a single C60 molecule. This quenching process appears to be catalytic. In other words the fullerene can react with many superoxides without being consumed. Due to this feature fullerenes are considered to be the world’s most efficient radical scavenger and are described as radical sponges (Krusic et al 1991).

Multiple studies have shown that fullerenes localize into cells, organelles like mitochondria and throughout extracellular spaces (HSDB, 2013). This allows for the carbon 60 to act as a reactive oxygen sink at a local level where the oxygen radicals are produced by cellular respiration, chemical exposure, or environmental exposure. The wide distribution of the product is a key to the successful antioxidant activity that carbon 60 provides. A major advantage of using fullerenes as biological antioxidant is their ability to localize within the cell to mitochondria and other cell compartment sites, where the production of free radicals takes place. Experiments on rats done by Najla Gharbi and coworkers proved this remarkable trait. They showed that aqueous C60 suspensions prepared without using any polar organic solvent, not only have no acute or subacute toxicity in rodents, but also protect their livers against free-radical damage (Gharbi et al 2005).”

One of the biologically most relevant features of C60 is the ability to quench various free radicals, behaving as a "free radical sponge". Conversely, photosensitization of C60 leads to its transition to a long-lived triplet excited state and the subsequent energy or electron transfer to molecular oxygen, yielding highly reactive singlet oxygen (1O2) or superoxide anion (O2-), respectively. These reactive oxygen species (ROS) react with a wide range of biological targets and are known to be involved in both cellular signaling and cell damage. Therefore, the dual property of fullerenes to either quench or generate cell-damaging ROS could be potentially exploited for their development as cytoprotective or cytotoxic anticancer/antimicrobial agents. However, the attempts to that effect have been hampered by the extremely low water solubility of C60, and by the fact that procedures to get C60 into solution profoundly influence the ROS-generating/quenching properties of C60 (HSDB, 2013).

Absorption, distribution and excretion strongly depend on the properties of the side chains on the fullerene. The number of pustules, consisting of accumulation of neutrophils, was significantly (P < 0.05) decreased from 1.45 +/- 1.13 to 0.18 +/- 0.60 (reduction rate 87.6%) (HSDB, 2013). Therefore, the fullerene gel may help in controlling acne vulgaris with skin care benefit (HSDB, 2013; Inui et al., 2011).
